Claudin-4 (CLDN4) messenger RNA is the transcript responsible for encoding the Claudin-4 protein, a key component of tight junction complexes that regulate paracellular permeability and maintain epithelial cell polarity [1, 2]. In various epithelial malignancies, including pancreatic, ovarian, and gastric cancers, CLDN4 mRNA is frequently overexpressed, contributing to tumor progression, cell survival, and chemoresistance [3, 4]. As a therapeutic target, CLDN4 mRNA is approached using RNA-based technologies such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs) to silence gene expression and reduce protein levels [5, 6]. This strategy aims to disrupt the tight junction barriers in tumors, potentially enhancing drug delivery and inhibiting metastatic pathways [7]. While protein-targeting agents like monoclonal antibodies are more clinically advanced, mRNA-targeted strategies offer high specificity for the CLDN4 sequence [8].
