Overview

Clostridium chauvoei toxin A (CctA) is the primary protective antigen and a critical virulence factor produced by the Gram-positive bacterium Clostridium chauvoei, which causes blackleg in cattle and sheep (Frey et al., 2012). As a member of the beta-barrel pore-forming toxin family, CctA functions by creating pores in the membranes of host cells, particularly leukocytes and erythrocytes, leading to cell lysis and extensive tissue necrosis (Frey & Falquet, 2015). This process results in the characteristic myonecrosis and rapid systemic toxemia that often prove fatal within 12 to 48 hours (Merck Veterinary Manual, 2023). In the pharmaceutical industry, CctA is the central component of clostridial bacterin-toxoid vaccines, which are designed to elicit a robust humoral immune response (Uzal et al., 2015). The resulting neutralizing antibodies bind to the toxin, preventing its interaction with host cell surfaces and thereby providing effective prophylaxis against infection (Abreu et al., 2017). While the antigen is highly effective for prevention, the rapid clinical course of the disease presents a significant challenge for therapeutic intervention using traditional antibiotics like penicillin (Rychener et al., 2017).
