The physical scaffold of the coagulation cascade refers to the membrane surfaces, primarily those provided by activated platelets or damaged endothelium, that serve as platforms for assembling coagulation factor complexes. Platelet membranes expose phosphatidylserine (PS), crucial for optimal fibrin formation. Fibrin itself, generated at the end stage of the cascade, forms a meshwork that stabilizes the platelet plug and supports cellular recruitment and tissue repair. Subendothelial matrix proteins exposed upon endothelial damage initiate platelet adhesion/activation. These scaffolds are essential for localizing enzymatic reactions, amplifying proteolytic cascades, stabilizing initial platelet plugs, and supporting subsequent tissue repair.
