The Fibrinogen / Thrombin system represents the terminal and most critical phase of the blood coagulation cascade. Thrombin (Coagulation factor IIa) is a potent serine protease that catalyzes the conversion of soluble fibrinogen (Coagulation factor I) into insoluble fibrin monomers, which subsequently polymerize to form the structural framework of a blood clot (StatPearls, 2023). Beyond its role in fibrin formation, thrombin also activates factors V, VIII, XI, and XIII, and stimulates platelet aggregation through protease-activated receptors (PARs), creating a powerful amplification loop for hemostasis (UniProt, 2024). Dysregulation of this system is a hallmark of various cardiovascular and hematological disorders, where excessive activity leads to arterial or venous thrombosis, while deficiency or inhibition results in significant bleeding risks (PubMed, 2022). Therapeutic strategies targeting this system include direct thrombin inhibitors (DTIs) like dabigatran and bivalirudin, which are used to treat and prevent thromboembolic events by blocking the enzymatic conversion of fibrinogen (NIH, 2023). Additionally, purified fibrinogen and thrombin are utilized clinically as topical hemostatic agents or "fibrin glues" to promote wound healing and control surgical bleeding (PubChem, 2024).
