Overview

Girdin, also known as Coiled-coil domain-containing protein 88A (CCDC88A), is a large scaffold protein that plays a pivotal role in regulating the actin cytoskeleton and cell motility [UniProt, 2024]. It functions as a non-receptor guanine nucleotide exchange factor (GEF) for Gαi subunits and is a critical substrate for the kinase Akt, which is essential for the migration of fibroblasts and cancer cells [PubMed, 2005; PubMed, 2009]. In many human malignancies, including glioblastoma and breast cancer, Girdin is overexpressed and serves as a driver of tumor invasion, metastasis, and angiogenesis [PubMed, 2017]. Consequently, Girdin mRNA has become a target for therapeutic intervention using RNA interference (RNAi) and antisense oligonucleotides (ASOs) to silence its expression and inhibit cancer progression [PubMed, 2012]. While these experimental therapies show promise in preclinical studies, challenges remain regarding the delivery of RNA-targeting agents and the potential for off-target effects on physiological processes like wound healing and vascular remodeling [PubMed, 2015]. Overall, Girdin represents a significant biomarker for cancer aggressiveness and a potential node for disrupting metastatic signaling pathways.
