Overview

Collagen alpha-2(IV) chain is one of six genetically distinct alpha chains that make up type IV collagen, which forms the major structural meshwork of basement membranes throughout the body[1][4][7]. Encoded by the COL4A2 gene, it pairs in a 2:1 ratio with the alpha-1(IV) chain (COL4A1) to form the predominant type IV collagen network. Beyond its architectural function, a C-terminal cleavage product known as canstatin possesses potent anti-angiogenic and anti-tumor properties by inhibiting endothelial cell proliferation, migration, and survival through integrin antagonism and induction of apoptotic pathways[3][4]. COL4A2 and the major type IV collagen network are critical to tissue integrity, filtration (notably in kidney glomeruli), cell signaling, and repair. Mutations in COL4A2 are associated with multiple diseases, including hereditary vascular syndromes, intracerebral hemorrhage, and other basement membrane pathologies[4][7].
