Overview

Collagen alpha-3(V) chain (COL5A3) encodes one of the alpha chains of type V collagen, a minor but functionally crucial fibrillar component of the extracellular matrix (ECM)[3][5][1]. Fibrillar collagen molecules, including type V, are trimeric structures important for the regulation of collagen fibril assembly in tissues that also contain type I collagen, thus influencing the structural integrity of connective tissue[3][5]. COL5A3 is structurally related to type XI collagen, and their chains may combine in tissue-specific manners. This protein has roles in cell–matrix interactions, wound healing, bone development, and metabolic homeostasis, with transient expression during tissue repair and adaptation to environmental stimuli such as hypoxia[1]. Mutations in COL5A3 have been implicated in a subset of Ehlers-Danlos syndrome, hypermobility type[3][5]. Disruption of COL5A3 can impair connective tissue architecture and has been associated with certain cancers and age-related tissue deterioration. However, COL5A3 is not commonly classified as a direct therapeutic target, receptor, enzyme, or transporter, and currently, there are no known drugs that specifically act through modulation of this protein[1][5][3].
