Colonic motility stimulation via increased stool volume is a physiological process involving a neuro-mechanical loop. Distension of the colonic wall by increased stool volume triggers serotonin release from enterochromaffin cells, activating sensory neurons and subsequently the enteric nervous system. This leads to coordinated smooth muscle contractions and relaxations (peristalsis) that propel colonic contents distally. While not a drug target *per se*, understanding this pathway informs therapeutic approaches to constipation and other motility disorders.
Increased stool volume leads to colonic distension, activating enterochromaffin cells (5-HT release). 5-HT stimulates sensory neurons via 5-HT4 receptors, initiating peristaltic reflexes through enteric nervous system (ENS) activation (acetylcholine/tachykinin release for contraction, nitric oxide for relaxation).
