Overview

Colorectal neoplasia differentially expressed (non-protein coding) (CRNDE) is a long non-coding RNA gene located on chromosome 16, expressed as multiple splice variants with highly tissue- and disease-specific patterns of expression. CRNDE was originally discovered as highly upregulated in colorectal cancer but is now recognized as one of the most upregulated lncRNAs in multiple cancer types, particularly glioma. CRNDE functions by interacting with chromatin-modifying complexes (e.g., Polycomb repressive complex 2/EZH2 and CoREST) to regulate the epigenetic landscape and promote oncogenic processes such as cell proliferation, migration, invasion, and inhibition of apoptosis in cancer cells. It may also act as a competing endogenous RNA, sequestering specific microRNAs, thereby impacting downstream gene expression. Upregulation of CRNDE is strongly correlated with tumor progression and malignancy in several cancers and may have roles in stem cell biology and neural development. No approved drugs currently target CRNDE directly, but it is considered a potential therapeutic target and prognostic biomarker in oncology
