Overview

Connective tissue growth factor (CCN2), also known as CCN family member 2, is a secreted matricellular protein that serves as a critical coordinator of the extracellular matrix (ECM) environment. It belongs to the CCN family and is characterized by a modular structure that enables interactions with a wide array of ligands, including integrins, heparan sulfate proteoglycans, and growth factors like TGF-beta and VEGF. Biologically, CCN2 is essential for processes such as cell adhesion, migration, proliferation, and the synthesis of ECM components, particularly during development and wound repair. However, its chronic overexpression is a hallmark of various fibrotic diseases, including idiopathic pulmonary fibrosis, systemic sclerosis, and diabetic nephropathy, where it drives excessive collagen deposition. In oncology, CCN2 is implicated in tumor growth, angiogenesis, and metastasis across several cancer types. Therapeutic targeting of CCN2, primarily through monoclonal antibodies like pamrevlumab, aims to disrupt these pathological signaling axes to treat fibrosis and malignancy.
