Overview

Core 2 beta-1,6-N-acetylglucosaminyltransferase (C2GnT) is an enzyme that catalyzes the transfer of N-acetylglucosamine in a beta-1,6 linkage to core 1 O-glycans on mucin-type glycoproteins, generating core 2 O-glycan structures[1][3][4]. Three main isoforms (C2GnT1, C2GnT2, C2GnT3) exist, which are widely expressed in mammalian tissues[1][5]. The enzyme's activity generates branched O-glycan structures involved in cell adhesion, immune cell trafficking, T-cell apoptosis, and modulation of cell signaling[1]. C2GnT-mediated glycosylation changes are observed in cancer progression, immune disorders, and certain infections[3]. No direct drugs are known to target C2GnT, but its activity influences cell surface glycan structures relevant for selectin-ligand interactions, tumor biomarkers, and immune regulation[1][3][4].
