Overview

The Crimean-Congo hemorrhagic fever virus (CCHFV) glycoprotein precursor (GPC) is a polyprotein encoded by the viral M segment that is essential for viral assembly and entry (UniProt P21698). It is post-translationally processed by host proteases, including site-1 protease (S1P) and furin, into mature glycoproteins Gn and Gc, as well as non-structural proteins like NSm and GP38 (Zivcec et al., 2016, PLoS Negl Trop Dis). Gn and Gc form heterodimers on the viral envelope and are responsible for binding to host receptors and mediating pH-dependent membrane fusion (Garrison et al., 2007, Virology). Because they are the primary surface proteins, they are the main targets for neutralizing antibodies and T-cell mediated immunity, making them the central focus for vaccine candidates and therapeutic monoclonal antibodies (Fels et al., 2021, Science). Currently, there are no FDA-approved vaccines or specific therapeutics targeting CCHFV GPC, though several candidates like ADI-36121 are in preclinical or early clinical evaluation (Mishra et al., 2020, Cell).
