Overview

Cyclic AMP-responsive element-binding protein 1 (CREB1) mRNA is the messenger RNA transcript that encodes the CREB1 transcription factor, a pivotal regulator of gene expression in response to various signaling pathways (UniProt P16220). The protein translated from this mRNA binds to the cAMP response element (CRE) within gene promoters, influencing processes such as cell survival, proliferation, and glucose metabolism (NCBI Gene ID: 1385). In oncology, CREB1 mRNA is frequently overexpressed in malignancies such as acute myeloid leukemia (AML) and non-small cell lung cancer, where it promotes tumor growth and resistance to apoptosis (PubMed: 28233892). In the central nervous system, the CREB1 pathway is essential for synaptic plasticity and the consolidation of long-term memory (PubMed: 15148310). Therapeutic strategies targeting CREB1 mRNA primarily involve antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs) designed to reduce protein levels in pathological states where CREB1 is hyperactive. However, because of its fundamental role in normal physiology, particularly in the brain, achieving tissue-specific delivery and avoiding cognitive side effects remain significant therapeutic challenges.
