In many oncogenic contexts, the p21 promoter is silenced or downregulated, facilitating unchecked cellular proliferation and survival. Pharmacological targeting of this region typically involves the use of histone deacetylase (HDAC) inhibitors, such as Vorinostat, which promote an open chromatin structure to reactivate p21 expression (Richon et al., 2000, PNAS).
