Overview

The cytochrome bc1 complex, also known as Complex III, is a critical component of the mitochondrial electron transport chain in Plasmodium species. This enzyme complex catalyzes the transfer of electrons from ubiquinol to cytochrome c and is essential for maintaining mitochondrial membrane potential and ATP production in the parasite. It is a validated target for antimalarial therapy, with drugs like atovaquone targeting the Qo site. However, resistance can develop rapidly through mutations. Newer inhibitors target either or both Qo and Qi sites with varying selectivity. Structural studies guide rational drug design efforts to overcome resistance while minimizing toxicity.
