Overview

Cytochrome c oxidase subunit 8C, mitochondrial (COX8C), is a nuclear-encoded protein subunit of cytochrome c oxidase (complex IV), the terminal enzyme of the mitochondrial electron transport chain[1][7]. Complex IV catalyzes the reduction of molecular oxygen to water, a critical step in oxidative phosphorylation that enables ATP production. COX8C is one of several nuclear-encoded, stoichiometric, non-catalytic subunits that participate in the assembly, stability, and possible regulation of cytochrome c oxidase, though it does not contribute directly to the active (catalytic) sites[1][4][7]. Mutations or deficiencies in complex IV subunits, including COX8C, can disrupt electron transfer, abolish oxidative phosphorylation, and cause severe mitochondrial diseases involving energy-deficient tissues such as brain, heart, and skeletal muscle[1][3][6][7]. There is currently no evidence for direct targeting of COX8C by drugs or use as a selective clinical biomarker.

Safety considerations

Dysfunction of COX8C or associated complex IV subunits leads to impaired energy metabolism, lactic acidosis, muscle weakness, heart, liver, and brain dysfunction[1][3][6].
