Overview

The cytokine receptor common subunit gamma (IL-2RG, also known as common gamma chain or CD132) is a shared signaling subunit of type I cytokine receptors for IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21. It forms heterodimeric or heterotrimeric receptor complexes with cytokine-specific subunits, associates with JAK1 and JAK3 kinases, and activates pathways including JAK-STAT, PI3K-Akt, and Ras-MAPK to regulate lymphocyte development, proliferation, differentiation, survival, and immune responses. Expressed on most lymphocytes, it is an integral membrane protein with extracellular, transmembrane, and intracellular domains, and its gene is on the X chromosome. Mutations lead to X-linked severe combined immunodeficiency (XSCID).
