Overview

The Interleukin-2 receptor common gamma chain (IL-2RG), also known as CD132, is a vital transmembrane glycoprotein that serves as a shared signaling component for several type I cytokine receptors, including those for IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21 (UniProt P34902). In mice, it is encoded by the Il2rg gene and is indispensable for the development and maintenance of the adaptive immune system, particularly for the maturation of T cells and Natural Killer (NK) cells (NCBI Gene 16186). Mutations in this gene lead to X-linked Severe Combined Immunodeficiency (X-SCID), a condition characterized by a near-complete absence of functional immune cells and profound susceptibility to infections (PubMed 15123770). From a therapeutic perspective, IL-2RG is a primary target for gene therapy approaches aimed at restoring immune function in SCID patients using viral vectors (PubMed 21148533). The receptor functions by associating with Janus kinase 3 (JAK3) to initiate downstream signaling through the STAT5 pathway, making it a focal point for research into immunomodulatory therapies for autoimmune diseases and transplant rejection (PubMed 8383671). Its central role in multiple cytokine pathways makes it a critical molecule for both basic immunology and clinical intervention strategies.
