Overview

DAPK1 intronic transcript 1 (DAPK1-IT1) is a long non-coding RNA transcribed from the intronic regions of the DAPK1 (Death Associated Protein Kinase 1) gene. Unlike protein-coding splice variants of DAPK1, DAPK1-IT1 functions at the RNA level, regulating DAPK1 mRNA stability and, as a competing endogenous RNA, modulating the availability of specific microRNAs such as miR-590-3p. Through these actions, DAPK1-IT1 can repress DAPK1 protein levels and modify cell migration and invasion, particularly in cancer cells, and also influence inflammatory processes relevant to atherogenesis. It does not function as a therapeutic target in the traditional sense (e.g., receptor, enzyme) but represents a regulatory node that could have biomarker or investigational therapeutic significance in cancer, cardiovascular, and inflammatory diseases[2][4][5].

Mechanism of action

Acts as a competing endogenous RNA (ceRNA) by sponging microRNAs such as miR-590-3p; also modulates DAPK1 mRNA stability, thereby influencing DAPK1 expression/function.

03

Biological functions

Regulation of mRNA stabilityRegulation of cholesterol metabolism and inflammatory response in macrophages (via miR-590-3p sponging)Regulation of gene expression, post-transcriptional modulation
04

Disease associations

Cancer (affecting tumor cell migration and invasion by regulating DAPK1 expression)Atherogenesis/cardiovascular disease (promotion of atherogenesis via cholesterol metabolism and inflammation)Asthma-related immune modulation
05

Biomarkers

Potential: Expression of DAPK1-IT1 could serve as a biomarker for diseases with disrupted DAPK1 signaling, tumor progression, or atherogenesis, but use is exploratory rather than established
