Target intelligence / Profile preview

DARKFOX (alternative open reading frame of FOXM1) (DARKFOX)

Target
DARKFOX
Molecular classification
Polypeptide product of an alternative open reading frame (ORF) of FOXM1, Tumor antigen (HLA-presented peptide antigen), Cancer antigen
01

Overview

DARKFOX is a novel, cancer-specific antigen derived from an alternative open reading frame (ORF) of the FOXM1 gene, robustly expressed at the surface of multiple solid tumor cell types and minimally in normal cells. It is the founding member of a new class of 'Dark Antigens,' discovered by Enara Bio, and represents a highly tumor-specific and homogeneously expressed target suitable for immunotherapies including bispecific T cell engagers. ENA101 is a lead therapeutic developed to specifically target DARKFOX, demonstrating potent cytotoxicity and strong potential to transform care for patients with diverse, high-prevalence solid tumors.

02

Mechanism of action

T cell-mediated cytotoxicity via recognition of DARKFOX-derived HLA-presented peptide, activated by bispecific T cell engager (ENA101)

03

Biological functions

Tumor antigen presentationImmune recognition (by T cells)
04

Disease associations

CancerSolid malignancies (lung, breast, gastrointestinal, squamous cell carcinomas, head & neck, esophageal cancers)
05

Safety considerations

Potential off-tumor toxicity (minimized: DARKFOX described as having minimal expression in normal tissue)Immunogenicity of bispecific T cell engager
06

Interacting drugs

ENA101 (bispecific T cell engager targeting DARKFOX)
07

Biomarkers

Expression of DARKFOX in tumor tissue for patient selection (implied by its use as a target for T cell therapy)
