Overview

DBH antisense RNA 1 (DBH-AS1) is a long non-coding RNA of approximately 2 kilobases, residing on chromosome 9q34. It is upregulated in various tumor tissues, particularly in HCC and DLBCL, often in response to hepatitis B virus X protein (HBx) and repressed by p53. Mechanistically, DBH-AS1 promotes cell cycle progression by increasing expression of cyclins (e.g., CCND1, CCNE1) and cyclin-dependent kinases, while lowering cyclin-dependent kinase inhibitors (e.g., p16, p21, p27). Activation of the MAPK signaling pathway by DBH-AS1 accelerates tumor cell proliferation and survival, and also inhibits serum starvation-induced apoptosis. In DLBCL, DBH-AS1 interacts with RNA-binding protein BUD13 to stabilize fibronectin 1 (FN1) mRNA, aiding tumor progression. Its expression has been proposed as a potential therapeutic target and biomarker, though conflicting expression data in some HCC studies highlight the need for further research. Quercetin, a natural compound, has been shown to downregulate DBH-AS1 in vitro, supporting the concept of its targeting in cancer therapy.
