Overview

Death-associated protein kinase 1 (DAPK1) mRNA is the messenger RNA transcript that encodes the DAPK1 protein, a calcium/calmodulin-dependent serine/threonine kinase that serves as a pivotal regulator of multiple cell death pathways, including apoptosis, autophagy, and necroptosis (UniProt: P53355). In its physiological role, DAPK1 acts as a tumor suppressor, and the silencing of its mRNA through promoter hypermethylation is a frequent epigenetic event in various human malignancies, making it a significant biomarker for cancer diagnosis and prognosis (PubMed: 21546401). Conversely, the pathological upregulation or overactivation of DAPK1 is strongly associated with neuronal death in acute conditions like ischemic stroke and chronic neurodegenerative diseases such as Alzheimer's and Parkinson's (PubMed: 30250274). As a result, DAPK1 mRNA is a target for experimental RNA-based therapeutics, including small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs), which aim to reduce DAPK1 protein levels to provide neuroprotection (PubMed: 24501107). The development of these therapies requires careful consideration of tissue-specific delivery and the potential risk of promoting tumorigenesis by inhibiting a known tumor suppressor. Current research focuses on optimizing the delivery of these RNA-targeting agents to the central nervous system while monitoring mRNA expression as a measure of therapeutic efficacy.
