Death effector domain-associated factor (DEDAF), also known as RING1 and YY1-binding protein (RYBP), is a multifunctional protein that plays a critical role in both the regulation of apoptosis and transcriptional repression (UniProt Q8N488). It was originally identified for its ability to interact with death effector domains (DEDs) of proteins like FADD and procaspase-8, thereby augmenting the formation of the death-inducing signaling complex (DISC) and enhancing apoptosis (Zheng et al., 2001). Beyond its role in cell death, DEDAF/RYBP is a core component of the non-canonical Polycomb Repressive Complex 1 (PRC1), where it facilitates chromatin remodeling and gene silencing essential for development and cellular differentiation (Gao et al., 2012). In oncology, DEDAF is frequently found to be downregulated in various malignancies, including lung, breast, and prostate cancers, suggesting a tumor-suppressive function (Garcia et al., 2008). DEDAF also interacts with the viral protein Apoptin, which contributes to its tumor-selective pro-apoptotic activity (Danen-van Oorschot et al., 2004). While there are currently no approved drugs specifically targeting DEDAF, its involvement in epigenetic regulation and apoptotic pathways makes it a subject of interest for therapeutic intervention in cancer and potentially neurodegenerative disorders (Wang et al., 2014).
