Overview

Deazaflavin-dependent nitroreductase (Ddn) is an enzyme found in *Mycobacterium tuberculosis* that activates nitroimidazole prodrugs like PA-824 (pretomanid), contributing to the treatment of tuberculosis. Ddn catalyzes the reduction of these compounds using the deazaflavin cofactor F420H2, leading to the intracellular release of reactive nitrogen species lethal to mycobacterial cells. Besides drug activation, Ddn also functions natively as an F420H2-dependent menaquinone reductase in mycobacterial electron transport chains, contributing to energy generation and oxidative stress defense.
