Overview

DLEU1 (Deleted in lymphocytic leukemia 1) is a long non-coding RNA located on chromosome 13q14.3, frequently deleted in hematopoietic malignancies such as chronic lymphocytic leukemia but is also found to be overexpressed and oncogenic in multiple solid tumors including breast, gastric, ovarian, endometrial, renal cell carcinoma, and oral squamous cell carcinoma[1][2][3][4]. DLEU1 acts as a regulator of cancer cell proliferation, migration, invasion, apoptosis resistance, and epithelial-mesenchymal transition through mechanisms involving competing endogenous RNA (miRNA sponging), transcriptional regulation, and modulation of key signaling cascades such as PI3K/Akt, ERK, and STAT3 pathways[1][2][4]. Experimentally, DLEU1 knockdown suppresses tumor growth, enhances apoptosis, and impairs metastatic behaviors of tumor cells. High expression of DLEU1 is correlated with poor prognosis in several cancers, and it is being explored as a potential biomarker and therapeutic target, although no drugs specifically targeting DLEU1 have been clinically approved to date[2][3][4].
