Overview

Dengue virus serotypes 1, 2, 3, and 4 are four closely related but antigenically distinct viruses in the Flavivirus genus, Flaviviridae family, with a roughly 11 kilobase positive-sense single-stranded RNA genome encoding three structural proteins (capsid [C], premembrane/membrane [prM/M], envelope [E]) and seven non-structural proteins (NS1, NS2A, NS2B, NS3, NS4A, NS4B, NS5)[4][1][3][6]. All four serotypes cause dengue fever in humans and, following infection, can lead to a spectrum of disease from mild febrile illness to severe, potentially fatal dengue hemorrhagic fever or dengue shock syndrome[1][4]. The envelope protein is the principal antigenic determinant, serving as the target for neutralizing antibodies and vaccines, but incomplete or cross-reactive immunity can lead to enhanced disease due to antibody-dependent enhancement; thus, vaccination and drug development face unique safety and efficacy challenges[2][4].
