Overview

The dengue virus NS5 protein is the largest and most conserved nonstructural protein encoded by the dengue virus genome. The N-terminal domain of NS5 possesses methyltransferase (MTase) activity, specifically functioning as a 2'-O-methyltransferase, which is essential for capping the viral RNA genome. This enzyme catalyzes methylation reactions required for forming the type I cap structure at the 5' end of viral RNA, protecting it from degradation, facilitating efficient translation, and evading host innate immune responses. It also interacts with host proteins to antagonize interferon signaling pathways. As a result of its crucial role in viral replication and immune evasion, the MTase activity of NS5 is an attractive target for antiviral drug development.
