The DNA intercalation site is a structural target within the deoxyribonucleic acid (DNA) double helix, specifically the space between adjacent base pairs (PubMed: PMC3135927). Planar, polycyclic aromatic molecules, known as intercalators, insert themselves into these sites through pi-stacking interactions, causing the DNA helix to unwind and lengthen (StatPearls: Anthracyclines). This structural distortion prevents the proper binding and movement of essential enzymes, including DNA and RNA polymerases and topoisomerase II, thereby inhibiting DNA replication and transcription (PubMed: 15013330). Intercalating agents like doxorubicin and dactinomycin are widely used in oncology to treat various leukemias, lymphomas, and solid tumors by inducing apoptosis in rapidly proliferating cells (NIH: NCI Drug Dictionary). Despite their efficacy, these drugs are associated with significant safety concerns, such as cumulative cardiotoxicity and myelosuppression, due to their relatively low selectivity for malignant over healthy tissues (PubMed: 28407941).
Intercalation involves the insertion of planar aromatic rings between DNA base pairs, leading to helix unwinding, inhibition of topoisomerase II, and disruption of DNA/RNA polymerase activity (PubMed: 15013330).
