Overview

DEP domain-containing protein 1 (DEPDC1) is a protein that plays a significant role in cell cycle progression and is frequently overexpressed in various malignancies, such as bladder cancer and non-small cell lung cancer (NSCLC) (Obara et al., 2012, Cancer Science). The HLA-A*24:02-restricted DEPDC1-derived epitope refers to a specific peptide sequence, typically DEPDC1-9-294 (EYYELGVHF), which is processed and presented by the HLA-A*24:02 allele on the surface of tumor cells (Takahashi et al., 2014, Journal of Immunotherapy). This peptide-MHC complex serves as a critical recognition site for cytotoxic T lymphocytes (CTLs), which can identify and lyse cells expressing the antigen (Obara et al., 2012, Cancer Science). Because DEPDC1 expression is highly restricted in normal tissues but significantly elevated in tumors, it is considered an ideal candidate for cancer immunotherapy (Yuan et al., 2014, World Journal of Surgical Oncology). Therapeutic interventions, such as the peptide vaccine OCV-C01, are designed to induce a robust, tumor-specific immune response against cells presenting this epitope (Takahashi et al., 2014, Journal of Immunotherapy). Clinical studies have demonstrated that targeting this epitope can lead to the induction of peptide-specific CTLs and potential clinical benefits in patients with HLA-A*24:02-positive cancers.
