Dermatophagoides pteronyssinus allergen 1-specific T-cell receptors (Der p 1-specific TCRs) are specialized protein complexes on the surface of T lymphocytes that recognize peptide epitopes from the major house dust mite allergen Der p 1 (UniProt P08176). These receptors play a pivotal role in the etiology of allergic asthma and rhinitis by mediating the recognition of allergens presented by MHC class II molecules on antigen-presenting cells (PubMed: 25652135). Upon binding, the TCR initiates a signaling cascade that typically promotes a Th2-biased immune response, characterized by the production of cytokines like IL-4 and IL-13, which drive IgE production and allergic inflammation (PubMed: 30102918). Therapeutically, these TCRs are the focus of allergen-specific immunotherapy (AIT), where repeated exposure to the allergen aims to desensitize the T cells or induce regulatory T cells (Tregs) to restore immune tolerance (PubMed: 28213130). Monitoring the frequency and phenotype of Der p 1-specific T cells via TCR sequencing or MHC tetramer staining serves as a valuable biomarker for treatment efficacy (PubMed: 24139497). Understanding the structural basis of the Der p 1-TCR interaction is also guiding the development of next-generation peptide vaccines and TCR-based diagnostics.
