The Dickkopf-related protein 3 (DKK3) mRNA 3' untranslated region (3'UTR) is a vital regulatory segment of the DKK3 transcript that controls protein expression through post-transcriptional mechanisms [UniProt, 2024]. This region contains multiple microRNA (miRNA) response elements (MREs) that serve as binding sites for various miRNAs, such as miR-125b, miR-425, and miR-92a [Oncotarget, 2017; J Cancer, 2020]. Upon binding, these miRNAs negatively regulate DKK3 expression by inducing mRNA degradation or inhibiting translation, thereby modulating downstream signaling pathways like the Wnt/beta-catenin axis [NIH, 2026]. DKK3 itself is a secreted glycoprotein involved in embryonic development and tissue homeostasis, often functioning as a tumor suppressor in various tissues [Wikipedia, 2024]. In many cancers, the downregulation of DKK3 via miRNA binding to its 3'UTR promotes tumor progression, invasion, and metastasis, making this region a significant focus for therapeutic intervention [MDPI, 2023]. Beyond oncology, the DKK3 mRNA 3'UTR is implicated in the regulation of renal fibrosis and acute kidney injury, where DKK3 levels in urine and serum serve as critical biomarkers for disease severity and progression [Frontiers, 2020]. Therapeutic strategies targeting the 3'UTR, including the use of miRNA mimics to restore DKK3 levels or antagomirs to block inhibitory miRNAs, are currently being explored to treat malignancies and fibrotic diseases [MDPI, 2023]. However, the context-dependent role of DKK3, which can act as an oncogene in certain stromal environments, presents a challenge for drug development [MDPI, 2023].
