Dietary phosphate refers to inorganic phosphate ingested through food, which is absorbed primarily in the small intestine. It is essential for numerous biological processes, including bone mineralization, energy metabolism, cell signaling, and nucleic acid synthesis. Absorption occurs via paracellular and transcellular pathways, regulated by vitamin D, dietary intake, and hormones. Targeting gastrointestinal phosphate absorption is clinically relevant in managing hyperphosphatemia, especially in chronic kidney disease.
Phosphate binders sequester dietary phosphate in the gut lumen, reducing its absorption. NHE3 inhibitors reduce paracellular phosphate permeability. NaPi-IIb inhibitors reduce transcellular phosphate uptake.
