Overview

Dietary triglycerides and related lipids represent the primary form of lipid energy consumed by humans, consisting of a glycerol backbone esterified with three fatty acids (National Center for Biotechnology Information, 2024). These molecules are essential for energy storage, insulation, and the absorption of fat-soluble vitamins, but their metabolism must be tightly regulated to maintain cardiovascular health (StatPearls, 2023). In the digestive tract, they are broken down by pancreatic lipases into smaller components for absorption, a process that can be therapeutically inhibited by drugs like Orlistat to manage obesity (Mayo Clinic, 2023). Elevated levels of these lipids in the blood, known as hypertriglyceridemia, are a major risk factor for atherosclerosis, metabolic syndrome, and acute pancreatitis (American Heart Association, 2021). Pharmacological management often involves the use of fibrates or omega-3 fatty acids, which lower triglyceride levels by increasing the activity of lipoprotein lipase or decreasing hepatic VLDL production (PubMed, 2022). While not a protein target itself, the regulation of dietary triglyceride levels is a central focus of metabolic and cardiovascular pharmacology.
