Overview

DIGIT (divergent to GSC, induced by TGF-beta family signaling), also known as GSC divergent transcript or GSC-DT, is a long noncoding RNA (lncRNA) induced by TGF-beta signaling during endodermal differentiation[1][5]. DIGIT is divergently transcribed from the GSC (Goosecoid homeobox) gene locus and is coordinately induced with GSC in embryonic stem cells undergoing differentiation[1]. It regulates endoderm differentiation by controlling GSC expression in trans, and DIGIT deficiency impairs this process; forced GSC expression can rescue the differentiation defect in DIGIT-deficient cells[1]. As a noncoding RNA, DIGIT does not encode a protein and is not considered a therapeutic target such as a receptor or enzyme. It functions at the regulatory level during development, primarily by modulating the expression of key lineage determinants in response to TGF-beta/SMAD3 signaling[1][5][3]. There is no evidence of direct association with drugs, clinical biomarkers, or established disease roles, though it may influence disease-relevant pathways such as TGF-beta signaling.
