Dihydroorotate dehydrogenase (quinone), mitochondrial (DHODH) is a flavoprotein enzyme located in the inner mitochondrial membrane. It catalyzes the fourth step in de novo pyrimidine biosynthesis, converting dihydroorotate to orotate using quinone as an electron acceptor. DHODH is essential for producing uridine monophosphate (UMP), a precursor for all pyrimidine nucleotides. Mutations in DHODH cause Miller syndrome. DHODH inhibition has been explored therapeutically for cancer, immunological disorders, and infectious diseases.
