Dimethylaminoethanol (DMAE), also known as deanol, is a naturally occurring amino alcohol and a structural analog of choline [1, 6]. It serves as a biochemical precursor to the neurotransmitter acetylcholine, which is essential for cognitive functions such as memory, learning, and attention [3, 9]. DMAE has been investigated for its potential in treating various neurological and behavioral disorders, including attention deficit hyperactivity disorder (ADHD) and Alzheimer's disease, although clinical evidence for its efficacy remains mixed [1, 5]. In addition to its role in neurotransmission, DMAE acts as an antioxidant and a cell membrane stabilizer, which has led to its widespread use in cosmeceuticals for skin firming and anti-aging [7, 8]. Despite its historical use as a prescription drug (Deaner), it is now primarily available as a dietary supplement [4, 6]. Safety concerns include potential cholinergic side effects, such as respiratory failure at high doses, and developmental risks, necessitating caution in its therapeutic application [1, 6].
Dimethylaminoethanol (DMAE) primarily functions as a precursor to choline and the neurotransmitter acetylcholine [1, 3]. It is thought to increase acetylcholine levels in the brain by crossing the blood-brain barrier more efficiently than choline and potentially inhibiting peripheral choline metabolism [2, 9]. Additionally, DMAE acts as a free radical scavenger and stabilizes cell membranes by being incorporated into phospholipids, and it has been shown to reduce the accumulation of lipofuscin in cells [7, 10].
