Overview

DIS3 like exosome 3'-5' exoribonuclease (DIS3L) is a cytoplasmic catalytic subunit of the multisubunit RNA exosome complex, exhibiting 3'-to-5' exoribonuclease activity that is essential for the degradation and processing of various RNA species. Unlike the related DIS3, which is primarily nuclear, DIS3L is restricted to the cytoplasm and collaborates with the exosome core to degrade unstable and normal mRNAs, thereby regulating cytoplasmic RNA turnover and protein synthesis. DIS3L plays a crucial role in embryonic development, with knockout studies demonstrating that its complete loss leads to embryonic lethality in mice. Mutations and dysregulation of DIS3L have been associated with cancer, including prostate cancer and Perlman syndrome. Despite its vital role in RNA metabolism, there are currently no known drugs directly targeting DIS3L, and its precise therapeutic utility remains under investigation[2][3][5].
