Overview

Carboplatin is a platinum-based chemotherapeutic agent that exerts its cytotoxic effects primarily through the formation of covalent adducts with DNA. The most significant of these are DNA crosslinks—both intrastrand and interstrand—which disrupt normal cellular processes such as replication and transcription, ultimately leading to cell death. Crosslinked DNA cannot be properly replicated or transcribed, triggering cell cycle arrest and apoptosis. Repair mechanisms exist but are often overwhelmed or defective in cancer cells. The extent and persistence of these lesions correlate with therapeutic efficacy as well as toxicity profiles.
