Overview

The N7 position of guanine in DNA is a highly nucleophilic site and the primary target for alkylation by endogenous and exogenous agents. Alkylation at this site leads to the formation of N7-guanine adducts, which are among the most common DNA lesions resulting from exposure to alkylating agents. While these lesions do not always directly miscode during replication, they can promote secondary lesions or strand breaks through depurination or further chemical reactions. Bulky modifications at guanine-N7 can impede nucleotide incorporation by some high-fidelity polymerases.
