DNA (platinum-induced crosslinking) refers to the covalent linkage of DNA strands or between DNA and proteins, mediated by platinum-based chemotherapeutic agents. These drugs form stable adducts with DNA, leading to cytotoxic effects that inhibit cell proliferation. The main adduct types are intrastrand crosslinks, interstrand crosslinks, monofunctional adducts, and protein-DNA-Pt ternary complexes. This crosslinking blocks replication/transcription and triggers apoptosis if unrepaired.
Platinum-based chemotherapeutics form covalent adducts with DNA, leading to crosslinking and distortion of the DNA double helix, inhibiting replication and transcription, and ultimately inducing apoptosis.
