Overview

Bendamustine is a bifunctional alkylating agent that forms covalent bonds with DNA bases through its electrophilic alkyl groups, resulting in intra-strand and inter-strand crosslinking. This DNA damage leads to inhibition of DNA replication and transcription, stem cell cycle arrest, and ultimately cell death via apoptosis and/or mitotic catastrophe. Bendamustine’s effects are distinct from other alkylating agents due to persistent DNA double-strand breaks and unique activation of DNA stress responses, contributing to its efficacy in various cancers and sometimes overcoming resistance seen with older agents. The phrase "DNA crosslinking via released bendamustine" thus refers to the mechanism whereby bendamustine, once administered and properly metabolized (released in its active form), directly crosslinks DNA, not to a discrete molecular target or receptor.
