Platinum-based agents induce DNA crosslinks, forming adducts primarily at guanine and adenine bases. These crosslinks, including intrastrand, interstrand, and DNA-protein crosslinks (DPCLs), distort the DNA helix, block replication and transcription, and ultimately trigger apoptosis. Repair pathways like NER and homologous recombination attempt to remove these lesions, but persistent DPCLs contribute to cytotoxicity.
