DNA crosslinking (alkylation) refers to the process of forming covalent linkages between two nucleotides within DNA, either on the same strand (intrastrand) or across complementary strands (interstrand). This is typically achieved through alkylating agents that transfer alkyl groups to specific positions on DNA bases. These crosslinks disrupt normal cellular processes such as replication and transcription, leading to cell death. It is a major mechanism of action for many chemotherapeutic drugs.
