DNA crosslinking, as induced by oxaliplatin, refers to the formation of covalent bonds between two nucleotides within DNA, either within the same strand (intrastrand) or between opposite strands (interstrand). Oxaliplatin is a platinum-based chemotherapeutic agent that exerts its cytotoxic effects primarily through this mechanism, leading to disruption of DNA replication and transcription, ultimately triggering cell cycle arrest and apoptosis. The bulky DACH ligand distinguishes oxaliplatin-induced DNA adducts from those formed by other platinum agents, influencing repair protein interactions and resistance profiles.
Forms DNA crosslinks (intrastrand and interstrand) primarily at guanine N7 positions, blocking replication and transcription.
