DNA crosslinking via platinum adduct formation is central to the mechanism by which several key chemotherapeutics exert cytotoxic effects against cancer cells. By forming stable intra/interstrand links—and occasionally ternary protein-DNA-platinum complexes—these drugs induce structural distortions that overwhelm cellular repair systems leading ultimately to cell death.
