DNA methyltransferase 1 (DNMT1) mRNA is the transcript responsible for the synthesis of the DNMT1 enzyme, the primary mediator of maintenance DNA methylation in mammals. This enzyme ensures the faithful inheritance of epigenetic marks by methylating hemimethylated CpG sites during the S-phase of the cell cycle (UniProt P26358). In various malignancies, DNMT1 is frequently upregulated, contributing to the epigenetic silencing of tumor suppressor genes and facilitating cancer progression (PubMed: 21880916). Therapeutic targeting of the DNMT1 mRNA, most notably through antisense oligonucleotides like MG98, aims to reduce protein expression levels to induce DNA hypomethylation and reactivate silenced genes (PubMed: 12467224). Beyond oncology, mutations in the DNMT1 gene are linked to neurodegenerative disorders such as hereditary sensory and autonomic neuropathy type 1E (PubMed: 21532572). Consequently, the DNMT1 mRNA serves as a critical node for therapeutic intervention in diseases characterized by epigenetic dysregulation.
Antisense oligonucleotide-mediated degradation of mRNA via RNase H recruitment, which prevents the translation of the DNMT1 protein and subsequently leads to DNA hypomethylation (PubMed: 12467224).
