Overview

DNA minor groove alkylation refers to the covalent modification of DNA bases within the minor groove of the double helix by alkylating agents. This process disrupts DNA replication and transcription, making it a target for anticancer drugs. Alkylating agents react with nucleophilic sites on DNA bases, particularly adenine N3, leading to cytotoxic lesions. The selectivity of alkylation can be enhanced using carrier molecules. The resulting damage triggers various DNA repair pathways, but if unrepaired or misrepaired, it can lead to mutagenesis and cell death. Minor-groove-directed alkylations are particularly effective at blocking RNA polymerase II-mediated transcription. This mechanism underlies the utility of alkylating agents as cytotoxic chemotherapeutics. Several repair pathways are involved, including Base Excision Repair (BER) and Nucleotide Excision Repair (NER).
