Overview

DNA polymerase alpha, delta, and gamma are three distinct enzymes essential for the replication and maintenance of eukaryotic genomes. DNA polymerase alpha (Pol α) initiates DNA replication by synthesizing RNA–DNA primers on single-stranded DNA[7]. DNA polymerase delta (Pol δ) is primarily responsible for lagging strand synthesis, also contributing to DNA repair, and functions with the accessory protein PCNA for processivity[6][7]. DNA polymerase gamma (Pol γ) is the primary polymerase for mitochondrial DNA replication and repair[2][3]. These enzymes, while sharing a basic catalytic mechanism, have non-redundant roles and distinct biochemical properties. Inhibiting any of these can disrupt DNA replication, making them both essential cellular targets and potential pharmacological targets for antiviral or anticancer therapies[7].
