DNA polymerase lambda (Pol λ), encoded by the POLL gene, is a member of the X family of DNA polymerases primarily involved in DNA repair [3, 5]. It plays a crucial role in non-homologous end joining (NHEJ) by filling gaps in double-strand breaks and serves as a key enzyme in base excision repair (BER) and translesion synthesis (TLS), particularly for bypassing oxidative lesions like 8-oxoguanine [1, 6, 15]. Pol λ also contributes to the diversification of the immune repertoire through its involvement in V(D)J recombination [3, 8]. In oncology, Pol λ is frequently overexpressed in tumors, which can lead to increased resistance to DNA-damaging chemotherapies [6, 9]. As a result, it is considered a promising therapeutic target; small-molecule inhibitors, such as rhodanine-based compounds, are being explored to enhance the efficacy of treatments like temozolomide by preventing the repair of drug-induced DNA lesions [4, 11].
