DnaJ heat shock protein family (Hsp40) member C15 (DNAJC15), also known as MCJ or DNAJD1, is a member of the highly conserved DnaJ/Hsp40 family of molecular chaperones and co-chaperones[3][6][7]. It acts as a negative regulator of the mitochondrial respiratory chain, restricting mitochondrial generation of ATP and preventing mitochondrial hyperpolarization[7]. DNAJC15 functions as an import component of the TIM23 translocase complex, stimulating the ATPase activity of HSPA9 and regulating the import of proteins into mitochondria[4]. It plays roles in regulating cell death pathways (apoptosis), and altered expression has been observed in cancer, suggesting its involvement in tumorigenesis and its potential as a biomarker[2][6]. DNAJC15 is not a receptor, enzyme, transporter, or transcription factor, but falls under the broader chaperone protein classification.
